چکیده مقاله:
Background Fertility preservation is an important part of scientific study in the field of reproductive medicine. Ovarian cryopreservation and in vitro follicles culture provide option for fertility conservation in older women or cancer patients. Basic fibroblast growth factor (bFGF) or FGF-2, is member of fibroblast growth factors family which play critical roles in cell migration proliferation, and differentiation during embryonic development. We planned this study to investigate the effect of Basic fibroblast growth factor in combination with follicular stimulating hormone on the follicular activation, maturation, viability and steroid production in human ovarian tissue culture after vitrification MaterialsAndMethods Ovarian tissue was obtained from (n=8) women undergoing gynecological operations. Then cortical tissue strips were vitrified for two weeks. After thawing cortical strip were divided to three groups including:FSH, FSH + bFGF (200 ng) and control (without FSH). Cortical strip were cultured in a (α-MEM) medium for 14 days. Then ovarian fragments were processed for histological and apoptosis status using Hematoxylin and eosin (H & E) staining and TUNEL kit respectively. Estradiol (E2), Anti mullerian hormone (AMH) and progestron concentration in culture medium were calculated using ELISA kit and viability were evaluated using Calcein-AM Ethidium homodimer-1. Results The percentage and diameter of primary and secondary follicles significantly increased in bFGF+ FSH group compared with other groups. Also Estradiol, progesterone and AMH production were higher in the bFGF + FSH group compared with other groups. In addition, the apoptosis status significantly decreased in bFGF + FSH group. Finally in FSH group the diameter of follicle increased and the apoptosis rate decreased compared with control group but it was not significant. Conclusion The bFGF-FSH combination activated primordial follicles and promotes growth of primordial and primary follicles in vitro. On the other hand bFGF-FSH decreases the rate of apoptosis during in vitro culture of human cortical strip.

متن کاملStimulation of ovarian stem cells by follicle stimulating hormone and basic fibroblast growth factor during cortical tissue culture
BACKGROUND Cryopreserved ovarian cortical tissue acts as a source of primordial follicles (PF) which can either be auto-transplanted or cultured in vitro to obtain mature oocytes. This offers a good opportunity to attain biological parenthood to individuals with gonadal insufficiency including cancer survivors. However, role of various intra- and extra-ovarian factors during PF growth initiatio...
متن کاملPioglitazone counteracts the tumor necrosis factor-α inhibition of follicle-stimulating hormone-induced follicular development and estradiol production in an in vitro mouse preantral follicle culture system
BACKGROUND Polycystic ovary syndrome (PCOS) is a common endocrine disorder in women of reproductive age and is characterized by chronic anovulation. Insulin resistance may be a key component of the pathogenesis of this disorder. Pioglitazone is a thiazolidinedione derivative that acts by improving insulin resistance via the peroxisome proliferator-activated receptor-γ (PPAR-γ) pathway. Reported...
متن کاملRole of Follicle Stimulating Hormone in the Survival, Activation and Further Growth of in vitro Cultured Sheep Primordial Follicles
The aim of the present study was to investigate the effect of follicle stimulating hormone (FSH) on survival, activation and growth of ovine primordial follicles using histological studies. Pieces of ovine ovarian cortex were obtained and cultured for 6 days in Minimum Essential Medium supplemented with or without FSH (50 ng/mL). These fragments were then process to be used for histology compar...

Follicle-stimulating hormone from different species has been used to increase ovulation and calving rates in cattle, injections proving to be most successful when given during the late follicular phase of the oestrous cycle or in combination with progestagens (Willett & Buckner, 1953; Bellows, Anderson & Short 1969). Some investigations have resulted in substantial increases in ovulation and ca...
متن کاملFollicle stimulating hormone increases spermatogonial stem cell colonization during in vitro co-culture
The complex process of spermatogenesis is regulated by various factors. Studies on spermatogonial stem cells (SCCs) have provided very important tool to improve herd genetic and different field. 0.2 to 0.3 percent of total cells of seminiferous tubules is consist of spermatogonial stem cells. To investigate and biomanipulation of these cells, proliferation and viability rate of cells should be ...
